Barum finished their Toolstation Western League Premier Division campaign second, but title-winners Odd Down withdrew their application to step up a tier.
Mill Road had already passed a Southern League inspection, but the club faced an anxious wait to see if they had been accepted into Division One South and West.
An FA meeting today – which has structured Step 1 to Step 4 in the non-league – confirmed Barry Yeo and his side will make the move up divisions.
Barnstaple are one of three teams to come up into the division, along with big-hitters Hereford FC and Salisbury.
They are joined by North Devon rivals Bideford and Paulton Rovers – both were relegated from the Premier Division.
The news caps off a memorable two years for Yeo, who has guided his side to back-to-back promotions and broken the club’s long-standing unbeaten record.
Next season Barnstaple will make trips to the likes trips to the likes of Taunton Town, Tiverton Town and Evesham United.
